I have a SATA HDD already filled with data and another SATA HDD that is blank. I don't want them to be mirrored, I would rather have the space. Can I put them both into DNS-323 without losing any information? Also will I be able to map the HDDs to my comptuer and access the file as if they are on another computer?

No, the DNS will repartition and format the disks, so better get a copy of your files before you put them into the NAS.
And yes - after having created shares and copied your files back to the DNS you can map the drives as if they are on another computer.

The DNS-323 uses the Linux EXT2 or EXT3 filesystem. Unless you are using Linux, you clearly can't stick that drive into the NAS and expect to find any data.

Could I put the blank HDD in first let that format, then transfer data, then put in the newly emptied HDD in to format?

Sure, don't see why not.
One thing that is clear here, you have no backup for this data. Obviously, the data isn't all that important, right? If it is important, I'd come up with a proper backup scheme.
