Is DNS Relay Enabled? How about Advanced DNS Service if you have this option?
Might need to create a Access Control rule as well as the Web Filter rule:

You might try using OPENDNS or a 3rd party DNS service, sign up with an account, use there DNS servers and use there web blocking or filtering options and see if blocking that site can be done there.

Is this one site or all sites that you input that fail to be blocked?

Try using Other Machines, the Block Some Access, check mark Apply Web Filter and Apply Advanced Port Filters, on the Next screen, you should see Dest IP Start and Dest IP End. Might try this. Not sure if this mean destination or local PCs though. if this doesn't work then you'll have to enter separate rules for each PC that your needed to block.  :-\
